ABSTRACT

ABSTRACT Purpose Quantify the tissue content of metalloproteinase-9 (MMP-9) and collagen in colic mucosa with and without intestinal transit after infliximab administration in rats subjected to Hartmann's surgery. Methods Twenty-two rats underwent colon diversion by Hartmann's surgery. Animals were maintained with intestinal bypass for 12 weeks to induce development of diversion colitis (DC). Afterwards, animals were divided into three groups: first group received subcutaneous application of saline solution (SS) 0.9%, while the remaining two groups received infliximab subcutaneously at doses of 5 or 10 mg·kg-1·week-1 for five consecutive weeks. After the intervention, animals were sacrificed, removing the segments with and without intestinal transit. Diversion colitis was diagnosed by histological study, and its intensity was determined by a validated inflammatory scale. Tissue expression of MMP-9 was assessed byimmunohistochemistry, while total collagen was assessed by histochemistry. Tissue content of both was measuredby computerized morphometry. Results Colon segments without intestinal transit had a higher degree of inflammation, which improved in animals treated with infliximab. Collagen content was always lower in those without intestinal transit. There was an increase in the collagen content in the colon without transit in animals treated with infliximab, primarily at a dose of 10 mg·kg-1·week-1. There was an increase in the content of MMP-9 in the colon without fecal transit, and a reduction was observed in animals treated with infliximab, regardless of the dose used. Conclusions Application of infliximab reduces inflammation, increases the total collagen content and decreases the content of MMP-9 in the colon without intestinal transit.

ABSTRACT

Resource utilization is an effective way to cope with the rapid increase of kitchen waste and excess sludge, and volatile fatty acids produced by anaerobic fermentation is an important way of recycling organic waste. However, the single substrate limits the efficient production of volatile fatty acids. In recent years, volatile fatty acids produced by anaerobic co-fermentation using different substrates has been widely studied and applied. In this paper, we analyze the characteristics of fermentation to produce acid using kitchen waste and excess sludge alone or mixture. Influences of environmental factors and microbial community structure on the type and yield of volatile fatty acids in the anaerobic fermentation system are discussed in detail. Moreover, we propose future research directions, to provide a reference for recycling kitchen waste and excess sludge.

ABSTRACT

The present study aimed to morphometrically evaluate the ruminal mucosa and ruminal fluid characteristics of Santa Inês sheep fed different levels of spineless cactus as a replacement for Tifton grass hay. A total of 32 intact male Santa Inês sheep, approximately 180 days old and with a mean weight of 32.0±1.6kg, were studied in a completely randomized design with four treatments and eight replicates. The morphometric measurements evaluated in the ruminal epithelium (papilla height and surface area) were significantly affected (P< 0.05) by supplementation of the diet with spineless cactus, and these effects showed increasing linear trends. Evaluations of the ruminal fluid also showed a significant increasing linear effect (P< 0.05) for ammonia and a quadratic effect for microbial protein. The inclusion of spineless cactus in the sheep diet affects the ruminal epithelium morphology, including the papilla height and surface area, and favors the absorption process in the rumen. The diet containing higher levels of spineless cactus led to maximum microbial protein production. These results characterize spineless cactus as a feasible alternative for feeding sheep during periods of drought, when the typical food source is scarce.(AU)


O presente estudo teve como objetivo avaliar morfometricamente a mucosa ruminal e as características do líquido ruminal de ovinos Santa Inês alimentados com diferentes níveis de palma forrageira como substituta do feno de capim-Tifton. Um total de 32 ovinos, machos inteiros, da raça Santa Inês, com aproximadamente 180 dias de idade e peso médio de 32,0±1,6kg, foi estudado em delineamento inteiramente ao acaso, com quatro tratamentos e oito repetições. As medidas morfométricas avaliadas no epitélio ruminal (altura e área superficial da papila) foram significativamente afetadas (P<0,05) pela suplementação da dieta com palma forrageira, e esses efeitos mostraram tendências lineares crescentes. As avaliações do fluido ruminal também mostraram um efeito linear crescente significativo (P<0,05) para a amônia e um efeito quadrático para a proteína microbiana. A inclusão de palma forrageira na dieta de ovinos afeta a morfologia do epitélio ruminal, englobando a altura e a área da papila, e favorece o processo de absorção no rúmen. A dieta contendo níveis mais altos de palma forrageira levou à produção máxima de proteína microbiana. Esses resultados caracterizam a palma forrageira como uma alternativa viável para a alimentação de ovelhas durante períodos de seca, quando a fonte típica de alimento é escassa.(AU)

Aim: This research aimed to study the ability of microbial consortium from buffalo feces to produce coal bed methane (CBM) in lignite coal through in-vitro technique.Methodology: This study was carried out in 2 stages: In first stage, microbes activated using in-vitro technique, and at second stage volatile fatty acids (VFA) and coal bed methane production were estimated. The nutrition source for inoculum activation was of three types: 100% concentrate: 70% concentrate: 30% grass and 30% concentrate: 70% grass. Biogas digester in laboratory-scale utilized 100 ml serum bottle filled with 70 ml, 98-5 media and 7 g coal. Microbial inoculum was inoculated on digester using a 10 ml syringe and incubated at 39oC. Parameters observed to measure the quality of inoculum were total number of anaerobic microbes, kinetics, and biogas production during fermentation. Complete Randomized Design with two factorial consisted of incubation period as factor A and inoculum dosages as factor B. Results: The activation process of inoculum from buffalo feces was required to produce coal bed methane in anaerobic digestion. During activation process, the microbes from buffalo feces in a mixture of 70% concentrate and 30% grass at 48 hr observation could produce total gas approximately 22.5 ml at 48 hr observation. Addition of activated buffalo feces anaerobic inoculum on the anaerobic digestion as much as 6% produced the highest number of anaerobic bacteria, and VFA accounted for 31 x 1010 CFU ml-1 and 171.7 mM, respectively. Meanwhile, the highest methane production reached 128.61 ml on adding 6% inoculum

BACKGROUND: Triclosan (TCS) is an antimicrobial agent widely used in health care and consumer products. This compound is present in sludge of wastewater treatment plants (WWTPs), and because of its bactericidal characteristics, it can inhibit the methanogenic activity in anaerobic digestion (AD) technology. The aim of this study was to evaluate the toxic effects of TCS on the methanogenic activity. RESULTS: Batch anaerobic reactors were used with TCS concentrations of 7.8, 15.7, 23.5, and 31.4 mg/L. These assays consisted in three successive feedings (I, II, and III), wherein the sludge was exposed to each TCS concentration and volatile fatty acid (VFA) substrate. For evaluation of the residual sludge activity during feeding III, only VFA was used. The results showed that the increase in TCS concentrations correlated with the reduction in methane (CH4) production. In this case, the minimum values were achieved for TCS concentration of 31.4 mg/L with CH4 levels between 101.9 and 245.3 during feedings I, II, and III. Regarding the effect of TCS on VFA consumption, an inhibitory effect was detected for TCS concentrations of 23.5 and 31.4 mg/L, with concentrations of acetic, butyric, and propionic acids at the end of the assay (37 d) between 153.6 and 206.8, 62.5 and 60.1, and 93.4 and 110 mg/L, respectively. Regarding the removal of TCS during AD, these values were above 47%. Conclusion: TCS is an inhibitor of methanogenic activity with a decrease between 63 and 70% during the different feedings. The CH4 production was not recovered during feeding III, with inhibition percentages of 21­72%.

ABSTRACT

Objetivou-se, com a presente pesquisa, avaliar a viabilidade de Aspergillus spp. com potencial probiótico durante o armazenamento e diante da pressão da microbiota autóctone, bem como a tolerância aos principais ácidos graxos da fermentação ruminal. Verificou-se também a inocuidade micotoxicológica desses isolados fúngicos cultivados em meio de cultivo sólido. Foram avaliados 20 isolados de Aspergillus spp., provenientes do trato gastrointestinal de bovinos criados em Urochloa decumbens lignificada. Esses fungos foram identificados por análise de sequências do DNAr e foram selecionados por apresentarem expressivo potencial celulolítico. O método vapor de amônia foi utilizado para detecção de cepas produtoras de micotoxinas. Os isolados foram avaliados quanto à viabilidade de crescimento em fluido ruminal por até 96 horas e estocagem em condições ambientais. Observou-se que os fungos avaliados não produziram aflatoxinas e que 95% dos isolados apresentaram resistência aos ácidos ruminais. Dois isolados, selecionados a partir das análises anteriores, apresentaram viabilidade sob a pressão da microbiota autóctone e de metabólitos do ecossistema ruminal e permanecem viáveis por, no mínimo, dois anos. Conclui-se que os isolados do gênero Aspergillus selecionados nesta pesquisa apresentam características fisiológicas para serem utilizados com aditivos microbianos ou probióticos para o ambiente ruminal.(AU)


The aims in this study were to evaluate the viability of Aspergillus spp. with probiotic potential during storage, pressure of autochthonous microbiota and tolerance to the main fatty acids of ruminal fermentation. The mycotoxicological safety was also verified. Twenty isolates from the gastrointestinal tract of cattle raised in lignified Urochloa decumbens were identified by rDNA sequence analysis and were previously selected because they showed significant cellulolytic potential. The ammonia vapor method was used to detect the production of mycotoxins. The isolates were evaluated for viability of ruminal fluid growth for up to 96 hours and storage under environmental conditions. The evaluated fungi did not produce aflatoxins, and 95% of them had resistance to ruminal acids. Two isolates, selected according these tests, presented viability on autochthonous microbiota pressure and metabolites from the ruminal ecosystem and remain viable for at least two years. In this research, the selected Aspergillus spp. isolates present physiological characteristics to be used with microbial additives or probiotic.(AU)

ABSTRACT

The aim of the current study was to investigate biogenic amines and mycotoxins concentrations in baled silage (mainly Poaceae family grasses) prepared in organic and conventional farms and to relate these parameters to fermentative parameters. The mean dry matter (DM) content was 364.10±93.31 and 424.70±95.93g/kg in the silage from organic and conventional farms respectively. The silage samples from organic farms had 17.00% higher (P≤ 0.05) tyramine (TY) than the silage from conventional farms. Conventional farm samples were characterized by 46.00% higher histamine (HIS) (P≤ 0.05), 9.80% higher putrescine (PUT) (P≤ 0.05), 17.30% higher cadaverine (CAD) (P≤ 0.05). Aflatoxins (AFL) (total) and zearalenone (ZEN), T-2/HT-2 concentrations were higher respectively 16.00% (P≤ 0.05) and 13.40% (P≤ 0.05), 1.80% (P≤ 0.05) in the silage prepared in organic farms. Deoxynivalenol (DON) concentration was higher 42.40% (P≤ 0.05) in silage from conventional farms. Volatile fatty acids (VFA), lactic acid, ethanol, pH and ammonia nitrogen showed that the silage samples from organic and conventional farms were of good quality. Our study suggests differences in biogenic amine formation or mycotoxins content in silage from organic and conventional farming, but, overall, the measured values are too low to be relevant for animal health. Furthermore, these differences might as well be due to the difference in dry matter content and plant maturity between the organic and conventional silage samples.(AU)


O objetivo do presente estudo foi investigar concentrações de aminas biogênicas micotoxinas em silagem embalada (principalmente gramíneas da família Poaceae) preparada em fazendas orgânicas e convencionais e relacionar esses parâmetros a parâmetros fermentativos. A massa seca média (MS) foi 364,10±93,31 e 424,70±95,93g/kg na silagem de fazendas orgânicas e convencionais, respectivamente. As amostras de silagem de fazendas orgânicas tinham 17% a mais de tyramina (TY) (p≤ 0,05) que as de fazendas convencionais. As amostras de fazendas convencionais foram caracterizadas por histamina (HIS) 46,00% mais alta (P≤ 0,05), 9,80% putrecina (PUT) mais alta (P≤ 0,05), 17,30% de cadaverina (CAD) mais alta (P≤ 0,05). Aflatoxnas (AFL) (total) e zearalenone (ZEN), T-2/HT-2 tinham concentrações mais altas em respectivamente 16,00% (P≤ 0,05) e 13,40% (P≤ 0,05), 1,80% (P≤ 0,05) na silagem preparada em fazendas orgânicas. Deoxinivalenol (DON) tinha concentração mais alta 42,40% (P≤ 0,05) na silagem de fazendas convencionais. Ácidos graxos voláteis (AGV), ácido lático, etanol, pH e nitrogênio de amônia mostraram que as amostras de silagem de fazendas orgânicas e convencionais tinham boa qualidade. Nosso estudo sugere diferenças na formação biogênica de amônia ou micotoxinas em silagem de fazendas orgânicas ou convencionais mas, em geral, os valores medidos foram muito baixos para serem relevantes à saúde animal. Ademais, essas diferenças podem ser devido à diferença na matéria sólida e maturidade da planta entre as amostras de silagem orgânica e convencional.(AU)

ABSTRACT

Background Anaerobic digestion is a technology applied successfully to converting organic matter into biogas. However, the presence of inhibitory compounds such as antibiotics can adversely affect methane production. The aim of this study is to evaluate the toxic effect of chlortetracycline hydrochloride (CLOR) on the methanogenic bacteria. In order to study the methanogenic toxicity of CLOR, different concentrations of CLOR (10, 50, 100, 200 mg L- 1) were evaluated by methanogenic toxicity assays using three feedings. Results Maximum methane production was obtained for the assays with 10 mg CLOR L- 1, the values obtained were 277 ± 4.07; 193 ± 11.31 and 166 ± 7.07 mL for the first, second and third feedings, respectively. The average values for acetic, propionic and butyric acid at start of the experiments were 2104 ± 139; 632 ± 7.6; 544 ± 26 mg L- 1, respectively. The VFA values obtained finally of the experiment were dependent on the evaluated antibiotic concentrations, indicating that the efficiency of methanogenesis is directly affected by the CLOR concentration. Conclusions CLOR is an effective methanogenic bacteria inhibitor. Moreover, the results show that CLOR has a bactericidal effect on methanogenic activity given that methane production did not recover during the third feeding. This study shows that the 50% inhibitory concentration (IC50) for methanogenic bacteria in 10 mg L- 1.

ABSTRACT

A rapid headspace gas chromatography tandem mass spectrometric ( HS-GC/MS ) method was established for the analysis of volatile fatty acids ( VFAs ) in the feces. Feces were suspended by 6%phosphoric acid aqueous solution (1:2 m/V) and sealed in the headspace bottle for HS-GC/MS analysis. The HS-GC/MS method was optimized as follows: agitator temperature ( temp. ):80 ℃, syringe temp.:80 ℃, sample incubation time: 30 min, injection: 1 mL without split-flow. The chromatographic separation was performed on a DB-FFAP capillary column (30m×0. 25 mm×0. 25 μm) with injection port temp.:250 ℃. The temperature program ( initial temp. at 50 ℃ within first 1 min, and raised to 200 ℃ by 10 ℃/min) was employed by fixing the flow of carrier gas (high purity helium) at 1. 0 mL/min. The electron energy at -70 eV for electron impact ( EI ) ionization, ion source temp.: 250 ℃, transfer line temp.:280 ℃, the voltage of electron multiplier at 0. 95 kV. The spectra were recorded in the range of m/z 33-200 for full scan. The established HS-GC/MS method could be applied to analyze VGAs in the feces from human and rat appropriately. There are nine VFAs identified in the feces from human, and eight VFAs detected in the feces from rat by retrieving the NIST library, comparing with the standards and analyzing the MS data. Furthermore, the relative percentage contents of acetic acid, propionic acid and butyric acid accounted for roughly 85% of all VFAs by area normalization. The method is simple and sensitive, and it can be used to rapidly detect VFAs in the feces from human and rat.

ABSTRACT

Ácidos graxos voláteis (AGV) são importantes compostos intermediários indicadores da digestão anaeróbia. Este artigo apresenta dados de validação de uma metodologia para análise de uma mistura de sete AGV (C1 a C5) em amostras ambientais por cromatografia líquida de alta eficiência (CLAE). Foi utilizada uma coluna de exclusão iônica a 55ºC, volume de injeção de 10 µL e fase móvel (0,01 mol.L-1 H2SO4) a 0,6 mL.min-1. Os critérios de validação foram: limites de detecção e de quantificação, que ficaram na faixa de 5,0 a 10 mg.L-1 e 15 a 30 mg.L-1, respectivamente; linearidade (comprovada para todos os ácidos); repetitividade e sensibilidade (muito boa para a maioria dos ácidos); efeito de matriz (efeito de supressão observado para o ácido isobutírico) e exatidão (85 a 104%). Sendo assim, as condições operacionais adotadas se mostraram válidas para a quantificação de AGV em efluentes de reatores anaeróbios.


Volatile fatty acids (VFA) are important intermediate compounds that somehow indicate the efficiency of anaerobic systems. This paper presents results of method validation for seven VFA (C1 to C5) analysis in environmental samples by high performance liquid chromatography (HPLC). It was used an ion exclusion column kept at 55ºC; injection volume of 10 μL and mobile phase (0.01 mol.L-1 H2SO4) at 0.6 mL.min-1. The validation criteria adopted were: limits of detection and quantification, that fell in the ranges of 5.0 -10 mg.L-1 and 15-30 mg.L-1, respectively; linearity (verified for all acids); repeatability and sensibility (very good for most acids); matrix effect (suppression effect observed for isobutyric acid) and accuracy (from 85 to 104%). Therefore, the operational conditions adopted seemed to be adequate and valid to measure VFA in samples from anaerobic reactors.

Vinte e oito bovinos Brangus foram usados para determinar o efeito da suplementação de cobre e selênio no desempenho e na fermentação ruminal. Os animais foram divididos em: 1) C(Controle) - sem a suplementação de cobre e selênio; 2) Se - 2mg Se/kg de matéria seca na forma de selenito de sódio; 3) Cu - 40mg Cu/kg de matéria seca na forma de sulfato de cobre; 4) Se/Cu - 2mg Se/kg de matéria seca na forma de selenito de sódio e 40mg Cu/kg de matéria seca na forma de sulfato de cobre. As pesagens dos animais foram feitas em intervalos de 28 dias, após jejum completo de 18 horas. Foram colhidas amostras de líquido ruminal para análises de ácidos graxos voláteis e pH. O ganho de peso diário aumentou com a suplementação de Se (P<0,05). A ingestão de matéria seca não foi alterada pelos tratamentos (P>0,05). Os animais submetidos à suplementação com Cu apresentaram menor pH ruminal quando comparado com a suplementação Se/Cu (P<0,05). Os animais suplementados com Se/Cu apresentaram maior proporção de ácido acético quando comparado com o controle (P<0,05). Para o ácido propiônico e butírico, não houve diferenças significativas (P>0,05) entre os tratamentos. Apesar do pouco efeito na fermentação ruminal, as suplementações de selênio, cobre e selênio/cobre proporcionaram uma melhor eficiência alimentar.


Twenty-eight Brangus cattle were used to determine the effect of copper and selenium supplementation on performance and rumen fermentation. The animals were divided in: 1) (Control) - without supplementation of copper and selenium; 2) Se - 2mg Se/kg of dry matter as sodium selenite; 3) Cu - 40mg Cu/kg of dry matter as copper sulphate; 4) Se/Cu - 2mg Se/kg of dry matter as sodium selenite and 40mg Cu/kg of dry matter as copper sulphate. Animal weighing was performed in 28 day intervals, after 18 hours of fasting. Samples of rumen liquid were collected for analysis of volatile fatty acid and pH. The daily weight gain increased with selenium supplementation (P<0.05). The dry matter intake was not altered by treatments (P>0.05). The animals that received copper supplementation had lower pH rumen compared with selenium/copper supplementation (P<0.05). The animals supplemented with selenium/copper had a high proportion of acetic acid when compared to control (P<0.05). For propionic and butyric acid, there was no difference (P>0.05) among treatments. Despite the little effect on rumen fermentation, the supplementations of selenium, copper and selenium/copper provided better feed efficiency.

ABSTRACT

The objective of this study was to evaluate Sargassum meal as feed for sheep through the measures of in vivo digestibility, dry matter degradability, pH, ammonia and volatile fatty acids in rumen. The Sargassum algae used in this experiment were collected at the end of spring, when they are more abundant, bigger, and have completed their reproductive cycle. Four tons (wet weigth) were collected manually from the intertidal zone of La Paz bay, Baja California Sur, Mexico. These algae were sun-dried and ground in a hammer mill to obtain the Sargassum meal. Four fistulated Pelibuey sheep, were fed daily with diets containing the marine algae (MA) at different levels (0, 10, 20 and 30 %), using a 4 x 4 Latin-square design experiment. Feed intake was not affected (p>0.05). Water consumption and urine excretion increased with MA (p<0.05; r²=0.54 and r²=0.74, respectively). In all treatments dry matter digestibility was of 74%-79%, and crude protein digestibility was of 85%-88%. Acid detergent fiber (59%-65%) and neutral detergent fiber (55%-66%) digestibility were greater in all treatments with MA. Ruminal pH was greater in all groups fed with MA (p<0.05). Ammonium concentration was not influenced (p>0.05) by MA. Ruminal volatile fatty acids decreased in all MA groups (p<0.05). The marine algae Sargassum spp. can be used as a feed supplement for sheep, especially in tropical and subtropical regions where these marine algae are available. Rev. Biol. Trop. 57 (4): 1271-1281. El objetivo de este estudio fue evaluar la harina del alga marina Sargassum como alimento para ovejas, midiendo la digestibilidad in vivo, la degradabilidad de la materia seca, así como el pH y los ácidos grasos volátiles en rumen. El alga Sargassum utilizada en este experimento, fue recolectada a finales de la primavera, cuando esta alga es más abundante, alcanza su mayor talla y ha completado su ciclo reproductivo. Se recolectaron manualmente, cuatro toneladas (peso húmedo) de la zona intermareal en la Bahía de La Paz, Baja California Sur, México. Estas algas fueron secadas directamente al sol y molidas en un molino de martillos, para obtener la harina. Se utilizaron cuatro borregos Pelibuey fistulados, distribuidos en un arreglo factorial de 4 x 4. Los animales fueron alimentados diariamente con dietas que contenían el alga marina (AM) Sargassum a diferentes niveles (0, 10, 20 y 30%). El consumo de alimento no se vio afectado con la inclusión del alga (p> 0.05). El consumo de agua y la excreción de orina se incrementaron conforme aumentó la concentración de AM en las dietas (p<0.05; r²=0.54 and r²=0.74, respectivamente). En todos los tratamientos la digestibilidad de la materia seca fue de 74% a 79%, la digestibilidad de la proteína cruda fue de 85% a 88%. La digestibilidad de la fibra ácido detergente (59%-65%) y de la neutro detergente (55%-66%) fue mayor en todos los tratamientos con AM, lo mismo ocurrió con el pH en rumen (p<0.05). La concentración de amonio en rumen no se vio afectada por AM (p>0.05). La concentración de ácidos grasos volátiles se redujo en todos los tratamientos con AM (p<0.05). El alga marina Sargassum spp. puede ser usada como complemento alimenticio para ovejas, especialmente en las regiones tropicales y subtropicales donde está disponible.

O trabalho investigou a influência do uso do extrato de levedura, fonte dos mediadores redox riboflavina e nicotinamida, na remoção de cor de solução de corante azo Drimaren Azul HF-RL em condições anaeróbias. O trabalho envolveu a execução de ensaios em batelada, em frascos-reatores mantidos a 25 ºC, incubados com o azo-corante e lodo anaeróbio na presença e ausência de fontes de carbono (extrato de levedura ou glicose) e de mediadores redox (riboflavina ou extrato de levedura). O monitoramento da variação temporal de cor, a demanda química de oxigênio (DQO) e ácidos graxos voláteis (AGV) mostraram que a adição de extrato de levedura (0,5 g/L) resultou em eficiências de remoção de cor de 80 a 85 por cento nas primeiras 24 horas de incubação, e que os produtos da degradação do azo-corante foram tóxicos para todo o consórcio anaeróbio, o que resultou em baixas eficiências de remoção de DQO na presença e ausência do extrato de levedura. Os resultados indicaram, ainda, que as eficiências de remoção de cor foram inferiores a 30 por cento na presença de apenas glicose (fonte de carbono) ou riboflavina (mediador redox), indicando que o extrato de levedura atuou simultaneamente como fonte de carbono e de mediadores redox.


This paper investigated the influence of using yeast extract, which is the source of redox mediators riboflavin and nicotinamide, in the decolorization of solutions containing the azo dye Drimaren Blue HF-RL in anaerobic conditions. It involved the incubation of serum bottles kept at 25 ºC and inoculated with the azo-dye, and anaerobic sludge in the presence and absence of carbon source (glucose or yeast extract) and redox mediators (riboflavin and yeast extract). The monitoring of color, chemical oxygen demand (COD) and volatile fatty acids (VFA) showed that the addition of yeast extract (0.5 g/L) resulted in 80 to 85 percent color removal in the first 24 hours of incubation; and that the metabolites of dye degradation were toxic to the anaerobic microorganisms, which led to low COD removal efficiencies either in the presence or absence of yeast extract. The results also showed that the efficiencies of color removal were below 30 percent in the presence of only glucose or riboflavin, indicating that the yeast extract acted simultaneously as source of carbon and redox mediators.

Dezessete bezerros foram utilizados para avaliar o efeito de ácidos graxos voláteis (AGV) sobre a morfologia ruminal, a epiderme do plano nasolabial, a epicera e o perioplum, e para validar a execução de biópsias tegumentares como indicadores de alterações da mucosa ruminal. Os animais receberam infusões intra-ruminal de butirato, propionato, lactato ou salina (controle) durante 37 dias. A insulina sorológica foi dosada no 22º dia experimental nos tempos de 0, 90, 180 e 360 minutos em relação à infusão diária da manhã. No 89º dia de vida, após o abate, foram coletados fragmentos ruminais e epidérmicos. Todos os AGV induziram aumento proporcionalmente maior no peso do ruminorretículo que no peso do omaso, sendo o butirato aparentemente mais estimulador da massa do estômago aglandular. Embora o butirato tenha sido mais estimulador da secreção de insulina, os AGV foram incapazes de induzir ganho nas dimensões papilares. Os AGV aumentaram a proliferação celular nos epitélios do rúmen e do perioplum traseiro, contrariamente ao efeito sobre o plano nasolabial e a epicera. Os efeitos dos AGV sobre a morfologia da mucosa ruminal e de outros tecidos queratinizados sugerem que danos morfológicos no rúmen e cascos podem ter causa comum. Biópsias tegumentares podem ter utilidade como indicadores de alterações morfológicas da mucosa ruminal.


The effect of butyrate, propionate, and lactate on ruminal wall, epidermis of nasolabial surface, perioplum, and epicera of 17 neonatal calves was evaluated. The experiment also aimed to validate the procedure and interpretation of tegument biopsies as indicators of ruminal mucosa alterations. Serum insulin was sampled on the 22nd day from the beginning of treatments, at 0, 90, 180, and 360 minutes after the morning infusion. After slaughtering, samples were collected from ruminal wall, nasolabial surface, epicera, and perioplum from face and hindquarters. All volatile fatty acids (VFA) induced greater increase in ruminal-reticulum proportionate weight than in omasum. Butyrate was a greater stimulator of non-glandular stomach growth. Although butyrate stimulated more insulin secretion, this VFA was not capable to induced gain on papillae area or height. The effect of nasolabial surface VFA infusion was the opposite to that observed in the rumen. VFA increased hind perioplum mitotic index, contrary to its effect on the nasolabial surface and epicera. VFA effects on ruminal mucosa morphology and on other keratinized tissues suggest that morphological damage on hoof and ruminal epithelium may have a common cause. Tegumentary biopsies may be useful as indicators of morphological alterations of ruminal mucosa.

Avaliou-se o efeito de ácidos graxos voláteis (AGV) sobre a integridade do epitélio no rúmen, no plano nasolabial, na epicera e no perioplum traseiro e dianteiro de bezerros e validou-se a feitura de biópsias tegumentares como indicadores de alterações morfológicas da mucosa ruminal. Dezessete bezerros, com sonda no rúmen, receberam infusões intra-ruminais de AGV ou salina, durante 37 dias. Aos 89 dias de vida, após o abate, foram colhidas amostras dos tecidos. Os AGV aumentaram a área de epitélio total e a área de células metabolicamente ativas no epitélio ruminal, embora o butirato não tenha induzido ao desenvolvimento papilar. A área de epitélio não queratinizado no plano nasolabial foi reduzida pela infusão de AGV. Butirato e lactato foram mais indutores de alterações patológicas no epitélio ruminal. Não foram observadas lesões histológicas nos epitélios do plano nasolabial, da epicera e do perioplum, mostrando que essas são conseqüências do efeito direto dos AGV sobre o epitélio ruminal. Os efeitos indireto e direto dos AGV sobre a morfologia dos tecidos epiteliais queratinizados não foram iguais. Biópsias tegumentares podem ter utilidade como indicadores de alterações morfológicas da mucosa ruminal.


The effect of volatile fatty acids (VFA) on rumen wall, epidermis of nasolabial surface, perioplum, and epicera of calves was evaluated. The experiment also aimed to validate the procedure of tegument biopsies as indicators of ruminal mucosa alterations. Seventeen neonatal calves with foley catheters received intraruminal infusions of VFA or saline, during 37 days. At 89-day-old, the animals were slaughtered and tissue samples were collected from rumen, nasolabial surface, epicera, and perioplum from face and hindquarters. VFA infusion increased total epithelium area and metabolically active ruminal cell area; although butirate did not induce the papilar development. The effect of nasolabial surface VFA infusion was the opposite to that observed in the rumen. No histological lesion was observed on nasolabial surface, epicera, and perioplum, demonstrating that these are consequences of VFA direct effect on ruminal epithelium. Butyrate and lactate induced more alterations on the ruminal epithelium. Indirect and direct VFA effects on keratinized epithelium tissues morphology were not identical. Tegumentary biopsies may be useful as indicators of morphological alterations of ruminal mucosa.

ABSTRACT

Sistemas de tratamento anaeróbios são sensíveis às mudanças de condições ambientais que influenciam no metabolismo dos microrganismos responsáveis pela estabilidade do processo. Os objetivos deste trabalho foram (i) avaliar a acurácia de métodos que determinam os parâmetros de controle ácidos graxos voláteis (AGV) e alcalinidade a bicarbonato (AB) em amostras preparadas com concentrações de 50 a 1000 mg/L e, (ii) verificar diferentes procedimentos de remoção de CO2 formado durante titulação ácida das amostras. A partir do teste estatístico de Tukey aplicado aos resultados obtidos, observou-se que os métodos Kapp e DiLallo & Albertson com utilização de ultra-som para a remoção de CO2 do meio líquido apresentaram os melhores resultados para AGV. Os métodos da Condutividade, Kapp e Jenkins se destacaram na determinação de AB.


Anaerobic treatment systems are sensitive to the environmental condition changes. Such changes may have influence on the metabolism of the microorganisms responsible for the process stability. The aims of this work were (i) to evaluate the accuracy of methods to determine control parameters volatile fatty acids (VFA) and bicarbonate alkalinity (BA) in samples prepared with concentrations from 50 to 1000 mg/L and, (ii) to verify different procedures of the CO2 removal formed during the acid titration of the samples. From the Tukey statistical test applied to the obtained results it was observed that the Kapp and DiLallo & Albertson methods using sonic equipment to remove CO2 from the liquid presented better results for VFA. The Conductivity, Kapp and Jenkins methods were considered the most reliable for BA determination.

ABSTRACT

Two continuous stirred tank reactors (CSTRs) each fed with palm oil mill effluent (POME), operated at 37ºC and 55ºC, respectively, were investigated for their performance under varies organic loading rates (OLRs). The 37ºC reactor operated successfully at a maximum OLR of 12.25 g[COD]/L/day and a hydraulic retention time (HRT) of 7 days. The 55ºC reactor operated successfully at the higher loading rate of 17.01 g[COD]/L/day and had a HRT of 5 days. The 37ºC reactor achieved a 71.10% reduction of chemical oxygen demand (COD), a biogas production rate of 3.73 L of gas/L[reactor]/day containing 71.04% methane, whereas the 55ºC reactor achieved a 70.32% reduction of COD, a biogas production rate of 4.66 L of gas/L[reactor]/day containing 69.53% methane. An OLR of 9.68 g[COD]/L/day, at a HRT of 7 days, was used to study the effects of changing the temperature by 3ºC increments. The reactor processes were reasonably stable during the increase from 37ºC to 43ºC and the decrease from 55ºC to 43ºC. When the temperature was increased from 37ºC to 46ºC, the total volatile fatty acid (TVFA) concentration and biogas production was 2,059 mg as acetic acid/L and 1.49 L of gas/L[reactor]/day at day 56, respectively. When the temperature was reduced from 55ºC to 40ºC, the TVFA concentration and biogas production was 2,368 mg as acetic acid/L and 2.01 L of gas/L[reactor]/day at day 102, respectively. By first reducing the OLR to 4.20 g[COD]/L/day then slowly increasing the OLR back to 9.68 g[COD]/L/day, both reactors were restored to stable conditions at 49ºC and 37ºC respectively. The initial 37ºC reactor became fully acclimatized at 55ºC with an efficiency similar to that when operated at the initial 37ºC whereas the 55ºC reactor also achieved stability at 37ºC but with a lower efficiency.

ABSTRACT

The composition of volatile fatty acids in the biogas digester based on cattle manure as substrate and stabilised at 25°C showed that it contained 87-88% branched chain fatty acids, comprising of isobutyric and isovaleric acids, in comparison to 38 % observed in the digester operating at 35°C. Mixed cellulolytic cultures equilibrated at 25°C (C-25) and 35°C (C-35) showed similar properties, but rates of hydrolysis were three times higher than that observed in a standard biogas digester. The proportion of isobutyric and isovaleric were drastically reduced when C- 25 was grown with glucose or filter paper as substrates. The volatile fatty acids recovered from C-25 (at 25°C) inhibited growth of methanogens on acetate, whereas that from C-35 was not inhibitory. The inhibitory effects were due to the branched chain fatty acids and were observed with isobutyric acid at concentrations as low as 50 ppm. Addition of another micro-organism Rhodotorula selected for growth on isobutyric completely reversed this inhibition. Results indicate that the aceticlastic methanogens are very sensitive to inhibition by branched chain fatty acids and reduction in methane formation in biogas digester at lower temperature may be due to this effect.
